RIASSUNTO
Scopo - Gli obiettivi dello studio sono stati quelli di validare un metodo automatizzato per la rilevazione dell’attività sierica di PON1 nella specie equina, basato sull’utilizzo del paraoxone come substrato e di determinare gli intervalli di riferimento (reference intervals, RIs) in cavalli e puledri sani.
Materiali e Metodi. Nel presente studio sono stati inclusi 120 cavalli e 55 puledri clinicamente sani. Su ogni soggetto è stato eseguito un prelievo di sangue e il siero è stato utilizzato per valutare l’attività di PON1 con metodo automatizzato e lettura spettrofotometrica utilizzando il paraoxone come substrato. Sono state valutate ripetibilità intra- e inter-assay, accuratezza tramite LUD (evaluation of linearity under dilution) e SRT (spiking recovery test). Sono stati determinati gli intervalli di riferimento in cavalli sani e analizzate le possibili differenze dipendenti da sesso, età e razza.
Risultati. I coefficienti di variazione risultati dagli studi intra- e inter-assay sono inferiori al 3% per tutte le condizioni di laboratorio esaminate. I test LUD e SRT hanno evidenziato un’ottima correlazione tra i valori attesi ed osservati. Nelle femmine adulte sono stati registrati valori più elevati rispetto ai maschi e ai castroni. Nei trottatori adulti sono stati registrati valori più elevati rispetto agli Warmbloods adulti.
Conclusioni. Il metodo automatizzato per la rilevazione dell’attività sierica di PON1 nella specie equina, basato sull’utilizzo del paraoxone come substrato è risultato accurato e preciso. Si suggerisce l’utilizzo di un unico intervallo di riferimento per giovani puledri e cavalli adulti, indipendentemente dal genere e razza.
ABSTRACT
Aim. The objectives of the study were to validate an automated paraoxon-based assay to measure PON1 activity in equine serum and to determine a reference interval (RI) in healthy horses and foals.
Materials and methods. 120 healthy adult horses and 55 healthy foals were enrolled in this study. Blood samples were collected and serum was harvested. Serum PON1 activity was measured spectrophotometrically using an automated analyzer through paraoxon-based method. Intra-assay and inter-assay imprecision, linearity under dilution (LUD) and SRT (spiking recovery test), PON1 RIs were determined for healthy horses and the possible differences depending on gender, age and breed were investigated
Results. Intra- and inter-assay CVs (coefficients of variation) were lower than 3% for all the laboratory conditions explored in this study. LUD and SRT tests fitted the linear model. RI recorded in the whole caseload was 38,1-80,8 U/mL. Adult females’ RI recorded was higher than males and geldings ones. In adults Trotters were recorded higher PON1 activity than in adults Warmbloods.
Discussion and conclusions. The automated paraoxon-based method to assess serum equine PON1 activity was accurate and precise. It would be advisable to use a single reference interval for young foals and adult horses, independently on their gender or aptitude.
