Ciclo biologico della vitamina D
Nel sangue la vit D circola legata con la Vitamin D Binding Protein (DBP) da cui è rilasciata al fegato, dove è idrossilata in posizione C-25 da una 25-idrossilasi (prob. cit. P450 o CYP2R1) a 25(OH)D (pro-ormone)
La 25(OH)D, viene legata dalla DBP, ha una emivita di 2 settimane e rappresenta la misura del contenuto di vit D dell’organismo
A livello del rene la 25(OH)D viene legata da un recettore di membrana (megalina) del tubulo e convertita da una 1-idrossilasi (CYP27B1) mitocondriale a 1,25(OH)2D (forma biologicamente attiva) regolata
dal PTH
La 1,25(OH)2D si lega al Vitamin D
Receptor (VDR) nel nucleo della cellula, e, successivamente, al Retinoic Acid X Receptor (RXR), formando un
complesso tri-molecolare che può così legare dei Vitamin D Responsive
Elements (VDRE), promuovendo la co-attivazione o co-repressione
