5.3.1 Inibizione enzimatica
L’attività inibitoria dei composti sintetizzati è stata saggiata addizionando 4 µL di una soluzione dell’inibitore alla miscela di reazione sopra descritta. Tutti i prodotti sono stati solubilizzati in DMSO 100% e diluiti alle concentrazioni appropriate con il tampone di reazione, fornito dal kit; la concentrazione finale di DMSO in soluzione non ha mai superato l’1% ed è stato dimostrato non influire sulla attività proteica. L’effetto inibitorio è stato estimato ad una concentrazione di 100 µM; i composti che sono stati scoperti essere attivi sono stati saggiati a concentrazioni addizionali, comprese tra 100 µM e 10 nM.
I valori di IC50, riportati in Tabella 17, sono stati ricavati mediante regressione non
lineare della curva del logaritmo della dose in funzione della risposta; tale curva è stata ottenuta utilizzando almeno 5 differenti concentrazioni dell’inibitore, in grado di determinare un’inibizione compresa tra il 20% e l’80%.

5.4 Conclusioni
I nuovi composti sintetizzati si sono rivelati essere efficienti inibitori tirosin- chinasici ad azione duale, in quanto attivi sia nei riguardi di VEGFR-2 sia della RET chinasi, come dimostrano i valori di IC50 dell’ordine del micromolare. In
particolare, il composto 2f, recante un atomo di fluoro in posizione para, ha mostrato il miglior profilo inibitorio (VEGFR-2, IC50=3.17 µM/RET, IC50=4.80
µM).
Ulteriori valutazioni funzionali verranno condotte dal dipartimento di Endocrinologia della Facoltà di Medicina, in collaborazione con il gruppo di ricerca presso cui ho svolto il mio lavoro di tesi.
