La somatotropina bovina ricombinante (rbST) è un ormone sintetico prodotto allo scopo di aumentare la resa delle vacche da latte. Se in alcuni Paesi il suo utilizzo è stato autorizzato, l’Unione Europea ne ha bandito il commercio e l’impiego all’interno dei suoi stati membri (Decisione del Consiglio 1999/879/CE).
L’estrema somiglianza fra la molecola della rbST e quella nativa ha reso finora infruttuosi i numerosi tentativi di sviluppare metodiche analitiche dirette per l’individuazione delle bovine trattate, stimolando la ricerca di approcci alternativi.
Questo lavoro di tesi s’inserisce in un progetto che si propone di analizzare l’espressione dei principali geni appartenenti all’asse somatotropo nella ghiandola mammaria e nel muscolo scheletrico di bovine trattate con rbST, con lo scopo di ricercare biomarcatori per la rivelazione del trattamento ormonale e di approfondire le conoscenze sui meccanismi molecolari di regolazione dell’asse stesso.
I risultati hanno rivelato un sensibile aumento d’espressione di IGFBP-5 in entrambi gli organi dopo trattamento con rbST, mentre sono risultati significativamente meno espressi
IGF-1R, IGF-2 e IGFBP-6 nella ghiandola mammaria e GHR nel muscolo, suggerendo un
coinvolgimento degli stessi nei meccanismi di regolazione dell’asse somatotropo. Tuttavia, le differenze osservate fra bovine trattate e controlli non possono essere considerate sufficienti a confermare un eventuale trattamento con rbST, a causa della loro limitata entità e delle variazioni individuali relative ai singoli animali.

The recombinant bovine somatotropin (rbST) is a synthetic hormone produced to increase the milk yield in cows. rbST is allowed in some Countries, yet the UE has banished its trade and use within the member States (Council Decision 99/218/CEE).
The little molecular difference between the recombinant and the native somatotropin has made fruitless the several attempts to develop direct analytic methods for the identification of treated cattle, thus stimulating the research of alternative approaches.
This work of thesis is part of a project the aim of which is to analyse the expression of the main genes of the somatotropic axis in mammary gland e skeletal muscle of dairy cows treated with rbST, with the purpose of researching biomarkers for detection of the hormonal treatment and of obtaining further insights into the molecular regulation mechanisms of the aforesaid axis.
The results show a considerable increase of IGFBP-5 expression in both organs after treatment with rbST, while IGF-1R, IGF-2 and IGFBP-6 in the mammary gland and GHR in the muscle result significantly less expressed, suggesting a possible involvement in the regulatory mechanisms of the somatotropic axis. Nevertheless, the differences observed between treated cows and control cannot be considered as a sufficient factor to confirm an eventual treatment with rbST, because of their limited extent and the individual variations with regard to the animals.
