ABSTRACT

This study is part of a research project oriented to analyse the changes of gene expression in brain structures involved in rats contextual fear learning. Studies of classical conditioning have produced a lot of information about emotional memory and associative learning of fear.

Classical fear conditioning is a form of learning that depends, mainly,

from amygdala activation. Whereas the contextual fear conditioning

(CFC) requires amygdala and hyppocampus activation, since the

environment where the animal receives the electric shock can produce

the response at the fear, conditioned at the contextual. Animals

submitted to CFC showed a typical behavioural response at the fear,

called “freezing”, every time that they are reinserted in the usual

apparatus of conditioning (retrieval test). Freezing behaviour last 4

weeks after the treatment. Electrophysiological recordings on

hippocampus slices of conditioned rats exhibited modifications of the

synaptic excitability along 7 days after the training. These long term

effects suggest that, after the CFC, a differential mRNA and new protein

synthesis occurs in the nervous system of rats. In order to identify the

genes involved we have utilized the Suppression Subtractive

Hybridisation technique (SSH) comparing, 48 h after the treatment, the

transcripts isolated from mid-brain of the CFC submitted rats to the

control rats, never entered in the conditioning apparatus. SSH generates

the isolation of sequences differentially expressed after treatment

through the construction of subtractive libraries of cDNA (starting from

poliA

mRNA extract of CFC and control rats). After evaluating the

efficiency of the subtraction, the cDNA has been cloned and undergoes screening. The differential clones, isolated with automatic sequencer, have been analysed and subsequently compared with the GeneBank, using the software FASTA and BLAST. Following bioinformatic analysis, the clones have been divided in functional category:

mitochondrial energy metabolism, creation of vesicles, mechanism of signal transduction and protein turn-over. The analysis of isolated genes expression has been done by means of RT-PCR real time. This technique, called quantity PCR or quantity PCR in real time (rtq-PCR) allows amplification and simultaneous quantification of nucleic material.

Our studies showed high homology for the genes coding the following

proteins: amphiphysin, caspase 3, p53 and stathmin.
