l'art. 36,comma 6, del D.Lgs. n. 50/2016 che prevede: "per l'affidamento e l'esecuzione di lavori, servizi e forniture di importo inferiore alle soglie di cui al precedente articolo 35, "Ie stazioni appaltanti possono procedere attraverso un mercato elettronico che consenta acquisti telematici basati su un sistema che attua procedure di scelta del contraente interamente gestite per via elettronica. Il Ministero dell'economia e delle finanze, avvalendosi di CONSIP S.p.A, mette a disposizione delle stazioni appaltanti il mercato elettronico delle pubbliche amministrazioni;

l'art. 15 ,comma 13, lettera d), della L.135/2012, così come modificato dalla L. 208/2015, che prevede: "fermo restando quanto previsto dall'articolo 17, comma 1, lettera a), del decreto legge 6 luglio 2011, n. 98, convertito con modificazioni dalla legge 15 luglio 2011, n. 111, gli Enti del Servizio Sanitario Nazionale, ovvero, per essi, le regioni e le province autonome di Trento e Bolzano, utilizzano, per l'acquisto di beni e servizi di importo pari o superiore a 1.000 euro, relativi alle categorie merceologiche presenti nella piattaforma CONSIP, gli strumenti di acquisto e negoziazione telematici messi a disposizione dalla stessa CONSIP, ovvero, se disponibili, dalle centrali di committenza regionali di riferimento costituite ai sensi dell'articolo 1, comma 455, della legge 27 dicembre 2006, n. 296. I contratti stipulati in violazione di quanto disposto dalla presente lettera sono nulli, costituiscono illecito disciplinare e sono causa di responsabilità amministrativa";

VISTI:

VISTO IL D. Lgs. 50/2016, recante il nuovo codice dei contratti pubblici relativi a lavori, servizi e forniture cosi come integrato dal D.Lgs19 Aprile 2017, n. 56;

VISTA la deliberazione N" 692 del 05/12/2016 avente ad oggetto "Semplificazione del procedimento amministrativo: ulteriori precisazioni";

VISTA la deliberazione N° 455 del 05/08/2016 avente ad oggetto "Affidamento dei contratti pubblici inferiori alla soglia comunitaria. Determinazioni di regolamentazione. Revoca delibera del Direttore Generale n. 621 del 24/12/2012";

VISTA la delibera n. 75 del 08.02.2017 "Rimodulazione articolazione interna del dipartimento amministrativo - presa d'atto nota dipartimentale n. 201700000151 del 04.01.2017. Provvedimenti conseguenti. "Con la quale, tra l'altro, viene ribadito che per l'effetto della nuova rimodulazione del Dipartimento Amministrativo l'Ing. Maria G. Lauletta, già Responsabile della UOSD Gestione Tecnico Patrimoniale ed incaricata ad Interim" della Struttura Organizzativa Provveditorato Economato, viene confermata nell'incarico di dirigente responsabile della rinominata "UOSD Gestione tecnico patrimoniale e Approvvigionamenti" conferito con delibera n. 506 del 08.09.2016 e con scadenza contrattuale confermata al 13.11.2019;

VISTA la delibera n.692 del 05.12.2016 "Semplificazione del procedimento amministrativo. Ulteriori precisazioni" con la quale, tra l'altro, in attesa della nuova articolazione organizzativa, la voce "U.O.

Provveditorato/Economato deve intendersi "Servizio Provveditorato/Economato;

VISTA la deliberazione n. 544 del 26.09.2016 con cui è stato conferito l'incarico ad interim della Struttura Organizzativa Provveditorato Economato all'ing. Maria Gelsomina LAULETTA, al fine di favorire l'integrazione delle risorse umane delle strutture e di utilizzare il personale in maniera condivisa;

5. Di dare atto che la documentazione inerente le procedure svolte restano custodite, anche in formato elettronico, presso il SERVIZIOProvveditorato/Economato;

4. Di affidare alle ditte citate la fornitura di materiale di consumo occorrente per le attività dei Laboratori di Ricerca di cui alle note Prot. W 12351 e W 12352 del 06/11/2017, e W 12590 del 09/11/2017 al costo totale di { 3.292,50 oltre Iva al 22% come per legge, a seguito delle procedure negoziate senza pubblicazione del bando, esplicate mediante Trattative Dirette con un solo operatore economico, ai sensi dell'art. 36 comma 2 lett. a) del Dlgs 50/2016 e del!' art. 9 del regolamento aziendale di cui alla delibera W 434 del 04/09/2015;

3. Di ratificare le Trattative W 307092 del 15/11/2017, W 312497 e W 312494 del 20/11/2017 ivi allegate;

2. Di richiamare integralmente la narrativa quale parte integrante e sostanziale del presente provvedimento;

The Seahorse XF Mito Fuel Flex Test determines the rate of oxidation of each fuel by measuring mitochondrial respiration [the oxygen consumption rate, (OCR) l of cells in the presence or absence of fuel pathway inhibitors (Figure 1 on page 6).

Sequentially inhibiting the pathway of interest followed by the two alternative pathways enables the calculation of how dependent the cells are on the pathway of interest to meet basal energy demand (Figure 2 on page 7). Dependency indicates that the cells' mitochondria are unable to compensate for the blocked pathway by oxidizing other fuels. Inhibiting the two alternative pathways followed by the pathway of interest enables the calculation of cells' mitochondrial capacity to meet energy demand (Figure 3 on page 8). Fuel Flexibility is

calculated by subtracting the Fuel Dependency from the Fuel Capacity for the pathway of interest (Figure 4 on page 9).

Flexibility indicates the cells' mitochondria have the ability to compensate for the inhibited pathway by using other pathways to fuel mitochondrial respiration. The presence of dependency and absence of flexibility demonstrates that the mitochondria require that fuel pathway to maintain basal OCR.

The Agilent Seahorse XF Mito Fuel Flex Test is a method for measuring mitochondrial fuel usage in live cells. In combination with the Agilent Seahorse XFe/XF96 or Agilent Seahorse XFe/XF24 Analyzer, the Agilent Seahorse XF Mito Fuel Flex Test Kit measures the dependency, capacity, and flexibility of cells to oxidize three mitochondrial fuels:

• Glucose (pyruvate)

• Glutamine (glutamate)

• Long-chain fatty acids.

Energyproduced by cells can be derived from mitochondrial oxidation of glucose. glutamine. and fatty acids. The cells' mitochondrial dependencyon and flexibility for each of these fuel sources is determined by measuring the decrease in fuel oxidation (decline in oxygen consumption rate) upon addition of one or more inhibitors.

The Seahorse XF Mito Fuel Flex Test Kit contains three pathway inhibitors required to determine the dependency, capacity, and flexibility of cells for glucose, glutamine and long chain fatty acids.

• UK5099 - An inhibitor of the glucose oxidation pathway.

UK5099 blocks the mitochondrial pyruvate carrier (MPC).

Cells convert glucose to pyruvate through glycolysis.

Pyruvate can be transported into the mitochondria and oxidized by the TCA cycle.

• BPTES - An inhibitor of the glutamine oxidation pathway.

BPTES is an allosteric inhibitor of glutaminase (GLSI).

Glutaminase converts glutamine to glutamate, glutamate is then converted to alpha-ketoglutarate, and oxidized by the TCA cycle.

• Etomoxir -

An inhibitor of long chain fatty acid oxidation.

Etomoxir inhibits carni ti ne palmitoyl-transferase lA

(CPTIA), which is criticai for translocating long chain fatty

acids from the cytosol into the mitochondria for beta

oxidation.

Fuel Dependency: Glutamine Oxidation Pathway Example Fuel Dependency is tested by first injecting an inhibitor of the target pathway, followed by inhibition of the two alternative pathways. Dependency is calculated using the equation shown here. In this example, HepG2cells were tested for Glutamine Pathway Dependency (n=3).BPTESwas injected following the third rate measurement (Baseline

Fuel Flexibility The difference between fuel capacity and dependency, that is, the ability of cells to increase oxidation of a particular fuel to compensate for inhibition of alternative fuel pathway(s).

Fuel Capacity The ability of a cell's mitochondria to oxidize a fuel when other fuel pathways are inhibited.

FuelDependency The measurement of cells' reliance on a particular fuel pathway to maintain baseline respiration.

Fuel Pathway A seri es of biochemical processes that convert fuels into metabolites that are oxidized in the mitochondria (example: the conversion of glucose to pyruvate and transport of pyruvate into mitochondria).

Fuel A substrate or nutrient that is used by cells and oxidized in the mitochondria. In this assay, the mitochondrial oxidation of glucose, glutamine, or long chain fatty acids is measured.

Baseline Respiration Rate of oxygen consumption due to fuel oxidation under initial assay conditions.

Prepare assay medium by supplementing Agilent Seahorse XF Base Medium. Seahorse recommends 1 mM pyruvate, 2 mM glutamine, and lO mM glucose as a starting point;

however, the desired medium composition can be varied depending on cell type or in vitro culture conditions. Refer to Basic Procedures for your analyzer.

2 Warm assay medium to 37 "C.

3 Adjust pH to 7.4 with 0.1 N NaOH (Note: sterile filtration following pH adjustment is recommended).

4 Keep assay medium at 37 "C until ready to use.

Important: Use compounds the same day they are reconstituted.

Do not refreeze. Discard any remaining compound. Refer to

"Removing reagent caps" on page 15 for instructions on removing the reagent caps.

Remove one foil pouch from Agilent Seahorse XF Mito Fuel Flex Test Kit box. Return box with remaining kits to -20°C.

2 Remove three tubes from the pouch, and place in a small tube rack. Allow the compounds to warm to room temp for approximately 15 minutes.

Resuspend the contents of each tube with prepared assay medium in volumes described in Table 3 with a pipette.

PIace cap on tube, and vortex for 1 minute to solubilize the compounds.

Remove the wash and add assay medium to total volume of 180 IlL (for Agilent Seahorse XF96 Microplates) or 500 IlL (for Agilent Seahorse XF24 Microplates). PIace the cell culture microplate into a 37°C non-C0

incubator for 1 hour prior to the assay.

Agilent Seahorse XFe/XF24: Compound preparation and sensor cartridge loading

For a Flexibility test, two groups are needed for each fuel of interest, one Dependency and one Capacity group.

Load compounds into the appropriate ports of a hydrated sensor cartridge (Refer to Tables 4-6). If using a template, follow the loading scheme provided on the Plate Map tab or redistribute groups appropriately (Refer to Figures 7-9).

Refer to the Basic Procedure for your analyzer for proper cartridge loading technique.

Of particular importance for the best quality data are items such as optimal cell density, consistent cell seedìng (as reflected in CVs of absolute baseline rates among similarly treated wells), and stable baseline OCR values of the cell line/type being used in the assay. For all XF assays, significant variability between wells within the same group indicates the need for additional optimization of cell culture, cell seeding, or

It

is also encouraged that absolute OCR values (in pmoljmin) of control and experimental groups are reviewed for any significant changes in OCR baseline values between groups (measurement 3/measurement prior to any injection). These differences in OCR values between groups suggest some biological change, and should be incorporated into the final interpretation of Mito Fuel Flex Test results.

Why is Dependency reported as zero?

If

Dependency is not significantly above zero or negative due to well to well variability, there is no dependency on that

particular substrate.

the cells are not dependent on the target fuel pathway, OCR may slightly increase following injection of inhibitor. When this occurs, Dependency is automatically set to zero (no dependence), and Flexibility will be equal to Capacity.

What does it mean if I have negative flexibility values?

When changes in OCR are small, well to well variability mìght lead to negative flexibility values. Negative flexibility values of less than 5 % are generally attributable to noise in the assay.

Other mitochondrial respiration: respiration dependent on an alternative substrate(s) being oxidized to support

mitochondrial respiration, which may include (but not limited to) short and medium chain fatty acids and amino acids other than glutamine.

Nonmitochondrial oxygen consumption: consumption of oxygen by other biochemical processes in the cell. This includes (but is not limited to) very long chain fatty acids that get partially oxidized in the peroxisomes and other cellular enzymatic processes that consume oxygen. The nonmitochondrial fraction of total oxygen consumption can be measured using the

Seahorse XF Cell Mito Stress Test.

Although not required, long chain fatty acid may be added to the medium. We recommend using a single species of long chain fatty acid, such as Seahorse XF Palmitate-BSA FAOSubstrate, when testing exogenous fatty acid oxidation. NOTE:only oxidation of long-chain fatty acid, such as palmitate, is sensitive to inhibition by etomoxir.

The recommended assay medium does not include fatty acid, can I add it?

Yes, the test uses all three compounds at concentrations well above their EC50 values for inhibition in mammalian cells.

These values have been validated in a variety of celIlines and primary isolates. While most cell types or celIlines have an appreciable response to at least one inhibitor, not all cells will respond to all inhibitors. If the cells are not responsive to a particular inhibitor, they may not be dependent on that

particular fuel pathway (that ìs, theyare flexible with respect to the fuel used for oxidative phosphorylation).

How do I interpret ECAR and glycolysis in this assay?

Using combinations ofinhibitors can confound interpretation of ECAR data with this test due to shifts in cellular ATP

production and demando For directly measuring glycolytic function, we recommend using the Seahorse XF Glycolysis Stress Test.

The above potential issues described (apparent low response to inhibitors,

dependency, or negative flexibility values) may be further diagnosed by performing (as a separate test or added group) a Mito Fuel Flex Test with only media injectìons (no inhibitors) to establish if baseline respiration changes significantly over the course of the assay. If the absolute respiration rates (OCR) in the absence of inhibitors trend significantly upward or downward throughout the assay, then the parameters of this test (relative

dependency, capacity and flexibility) will be either underestimated or exaggerated, respectively. This depends on the magnitude and direction of any baseline trends versus. the magnitude of change due to added inhibitors. To limit any upwardjdownward baseline trendìng OCR,ensure that cell culture and technical parameters of the assay have been thoroughly optimized.

The Agilent Seahorse XF Palmitate-BSA fatty acid oxidation (FAO) Substrate is

a bio-available palmitate reagent for researchers studyìng substrate utilization

who require a reliable source of conjugated fatty acids. The XF Palmitate-BSA

FAOSubstrate is manufactured at a 6:1 Palmitate:BSA ratio. Each lot is tested

Introduction

The ability to oxidize exogenously added fatty acids (FAs), as indicated byan increase in O

2

consumption rate (OCR), will be influenced by ATP demand, other substrates present in the assay medium (such as glucose), and stores of endogenous substrates (such as glycogen or triglycerides). Exogenously added FAs can also uncouple mitochondria, and cause an increase in the OCR not due to fatty acid oxidation (FAO).

Palmitate-BSA has been used extensively to examine FA oxidation. The addition of etomoxir to a cell, and the subsequent return to the control rate of respiration, demonstrate utilization of exogenous fatty acids

A drop in OCR in both the palmitate and BSA control indicates that endogenous FAs were also being oxìdìzed '. Finally, if the OCR signal is insensitive to etomoxir or oligomycin, this indicates that the FAs are uncoupling the mìtochondrìa", Because a change in OCR can be caused by utilization of exogenous FAs, endogenous FAs, or uncoupling by FAs, Agilent Technologies developed the XF FAOAssay that can measure all three sources. With the combined use of the XF Palmitate-BSA FAO Substrate, etomoxir, and the XF Cell Mito Stress Test, one can drive cells to oxidize exogenous fatty acids while revealing the portion of the OCR signal generated by endogenous and exogenous fatty acid

oxidation, as well as uncoupling.
