Elettroforesi Tricina-SDS-PAGE

E’ stata eseguita un’elettroforesi in tricina-SDS-PAGE delle frazioni 2, 3, 4 e 5, per separare le proteine da 26.6 kDa a 1.4 kDa. Come si può vedere in Figura 37, nella frazione 2 è presente una banda a 26.6 kDa dovuta a proteine ad alto peso molecolare, tuttavia sono presenti bande fino a un peso molecolare di circa 6.5 kDa; nelle frazioni 3 e 4 invece si ha una diminuzione di intensità delle bande presenti nella frazione 2, ma si ha un arricchimento di proteine con pesi molecolari compresi tra 6.5 kDa e 3.5 kDa. Va

74 sottolineato che, la precipitazione con TCA al 20%, non consente un recupero del 100% delle proteine presenti in ogni frazione, ma il recupero sarà circa del 70%.

Come atteso, invece, la frazione 5 non mostra alcuna banda, infatti in essa dovevano essere contenuti peptidi a basso peso molecolare, inferiori a 1.4 kDa, che non è possibile separare con questa tecnica.
